Fragment DNA
This step enzymatically fragments the DNA. An endpoint fragmentation is used to prevent overfragmentation.

| 1. | Preheat the heat block with the MIDI plate insert or the Illumina Hybridization Oven to 37°C. |
| 2. | Prepare the following consumable: |

Thaw at room temperature. Vortex or invert 10 times to mix. |
| 1. | Add 50 μl FMS to each well of the WGA plate. |
| 2. | Reseal with the cap mat using the original orientation. |
| 3. | Vortex at 1600 rpm for 1 minute, and then centrifuge at 280 × g for 1 minute. |
| 4. | Incubate on the preheated heat block or oven for 1 hour. |
Leave the plates on the heat block or oven until you have completed preparation for the next step. Do not leave the plates on the heat block or oven for longer than 2 hours.
| 5. | Centrifuge the plate(s) at 280 × g for 1 minute to remove condensation from the seal before storage. |
