Custom Kits

You can add custom or third-party index adapter and library prep kits to the MiSeq i100 Plus Control Software. The kits are available within the on-instrument Run Planning tool during run setup.

When adding a library prep kit, you must specify one or more compatible index adapter kits. If you need to add a custom index adapter kit, add it before adding the library prep kit.

Add a Custom Index Adapter Kit

1. Select the menu icon in the upper-left corner.
2. Select Settings, and then select Custom Kits.
3. Select Download Template to download the Index Adapter Kit template.tsv file
4. Open the template.tsv file using Microsoft Excel, Libre Office, or other similar spreadsheet editing software.
5. Follow the instructions in the template.tsv file to add the following index adapter kit information:
a. [IndexKit]—Overview information for the index adapter kit, including name, version, description, and index strategy.
b. [Resources]—Allows for providing adapter sequences for Read 1 and Read 2. Based on the values in this section, the imported file sets the index kit type as one of the following options:
Fixed layout single plate.
Fixed plate layout multi plate.
c. [Indices]— A list of indexes, including name, index sequence, and whether the index is for Index 1 or Index 2.

Index names can include only alphanumeric characters.

6. Remove the template instructions included in the angle brackets (< >), and then save the TSV file.
7. In the MiSeq i100 Plus Control Software user interface, select the drop-down menu in the upper left corner, and then select Custom Kits.
8. Select Import index adapter kit, then navigate to the custom index adapter kit *.tsv and select Open.
9. After successfully importing the custom index adapter kit, select the kit name to review and edit information.

Add a Custom Library Prep Kit

1. Select the menu icon in the upper-left corner.
2. Select Settings, and then select Custom Kits.
3. Select Add Library Prep Kit and enter the following information:
Library prep kit name.
[Optional] Description.
[Optional] Organization. The company or institution that owns the custom library prep kit. The organization cannot be Illumina.
Allowed read types.
Default read type.
Default read cycle.
From the drop-down list, select at least one compatible index adapter kit.
4. Select Save.
5. After successfully adding the library prep kit, select the kit name to review and edit information.
