Prepare for Protocol
This preparation is required to perform the protocol steps leading up to the overnight hybridization step. Illumina recommends that you gather and thaw reagents at the beginning of the day.

Thaw in a room temperature water bath (minimum 20 minutes). Make sure that the lid remains above water. Dry off before use. |
| 2. | Save the following Hyb program on the pre-PCR thermal cycler: |
| • | Choose the preheat lid option and set to 100°C |
| • | Set the reaction volume to 100 µl |
| • | 95°C for 10 minutes |
| • | Decrease by 0.1°C per second until reaching 50°C |
| • | 50°C for 1 hour |
| • | Decrease by 0.1°C per second until reaching 45°C |
| • | 45°C for 2 hours |
| • | Decrease by 0.1°C per second until reaching 40°C |
| • | 40°C for 4 hours |
| • | Decrease by 0.1°C per second until reaching 37°C |
| • | 37°C for 8 hours |
| • | Hold at 37°C |
