DNA Extraction Protocol

Due to the complex composition of urine samples, recommending a single DNA extraction kit or procedure that consistently extracts high quality DNA from a wide variety of organisms is difficult. The extraction protocol below is provided as a courtesy and has been tested and routinely used by Illumina in combination with this kit. Some reagents and materials must be purchased from non-authorized third-party suppliers, and Illumina does not guarantee or promise technical support for the performance of our products used with any of these reagents purchased from non-authorized third-party suppliers. The DNA extraction protocol is optimized to obtain successful DNA extraction for all targets within each pathogen class (ie, bacteria, viruses, fungi, and parasites) from urine in a single procedure. Complete the steps outlined in this section to extract DNA in preparation for sequencing. Reagents used in this extraction method are part of the Quick-DNA Urine Kit (Zymo Research, Catalog # D3061). We recommend purchasing the individual consumables as listed below instead of the complete Quick-DNA Urine Kit to ensure all required components are available and in appropriate quantity. Technical support for Extraction should be directed to Zymo Research.

The recommended sample input is 1.8 ml of urine. Different kits may have different sample recommendations. Sample backgrounds other than urine (such as saline solution, synthetic urine, and molecular biology grade water), are not guaranteed to yield successful DNA extractions.

To avoid cross-contamination, complete the DNA extraction protocol in an appropriate biosafety cabinet, following recommended aseptic techniques.
