If you are using enrichment probe panels from Illumina or third-party vendors, make sure that they meet the following specifications:

• 80 or 120 bp probe length
• Single- or double- stranded DNA probes
• 50 kb–2 Mb panel size for single-stranded probes
• 70 kb–2 Mb panel size for double-stranded probes

Illumina Custom Enrichment Panel v2

Prepare up to 32 samples using 4-plex enrichment, and up to 8 samples using 1-plex enrichment (32 μl, 120 bp)

Prepare up to 384 samples using 4-plex enrichment, and up to 96 samples using 1-plex enrichment (384 μl, 120 bp)

Prepare up to 1536 samples using 4-plex enrichment, and up to 384 samples using 1-plex enrichment (1536 μl, 120 bp)

Illumina Custom Enrichment Panel

Prepare up to 108 samples using 4-plex enrichment, and up to 27 samples using 1-plex enrichment

For index adapter sequences, refer to Illumina Adapter Sequences.

Each kit contains one plate of IDT for Illumina UMI DNA Index Anchors and one plate of IDT for Illumina DNA/RNA UD Indexes Set A or B. For Illumina UMI DNA/RNA UD v3 indexes, each kit contains one plate of IDT for Illumina UMI DNA Index Achors and one plate of Illumina DNA/RNA UD v3 indexes Set A and B.
