Dry Bath Operation
The protocol requires a dry bath with 0.5 ml tube block. Make sure that the appropriate block is installed.

Lid temperature is set manually and is independent from the dry bath programs. Understand how to configure lid mode and temperature before starting the protocol. |

The dry bath interface displays Lid On when the lid is off and Lid Off when the lid is on. |

For nuclei lysis and reverse transcription, use the specific temperature control mode to set the lid temperature to 105°C. When set to this mode, the lid temperature box shows the set value (for example, 105.0). |

For cell lysis, use the offset temperature control mode to set the lid temperature to 5°C higher than the heat block. When set to this mode, the lid temperature box shows the temperature offset (for example +5.0). |

To change the lid mode, select Edit, and then select LidMode to toggle between 105°C and +5.0, and then select Save/Return. |

The lid temperature only climbs to 105°C during incubation temperatures of 35°C or higher. |

The dry bath is preprogrammed with nuclei lysis and cell lysis incubation protocols. Do not modify the protocols. |
