Protocol

This section provides step-by-step instructions on how to prepare consumables, dilute libraries, and set up a sequencing run.

When handling reagents and other chemicals, wear safety glasses, a lab coat, and powder-free gloves.

Make sure that you have the required consumables and equipment before starting a protocol. Refer to Consumables and Equipment.

Follow the protocols in the order shown, using the specified volumes, temperatures, and durations.

You can initiate a sequencing run by selecting one of the following run types:

A planned run. Refer to Start a Planned Run.
A manual run that generates BCL files only. Refer to Start a Manual Run (Generate BCL Files).
A manual run that uses a sample sheet for local analysis. Refer to Start a Manual Run (Import Sample Sheet).

If analyzing data in the cloud, secondary analysis begins automatically in BaseSpace Sequence Hub or ICA. If analyzing data locally, on-instrument analysis begins automatically and output files are stored in the selected output folder.

If the storage is not sufficient to start a run, an error message prompts you to clear space.
