Initiate a Sequencing Run

You can initiate a sequencing run by selecting a planned run or by creating a manual run. To initiate a run on the idle side of the instrument while a run is in progress on the other side, refer to Staggered Start of Runs.

If analyzing data in the cloud, secondary analysis begins automatically in BaseSpace Sequence Hub or ICA. If analyzing data locally, on-instrument analysis begins automatically and output files are stored in the selected output folder.

Before starting a run, make sure that you have the necessary hard drive space available. Storage requirements are affected by the number of sequencing cycles, size of the samples, secondary analysis configuration, and compression type. The NovaSeq X Series can store at least 2 flow cells worth of 25B data for any secondary analysis configuration. For secondary analysis configurations that generate smaller amounts of data (for example, DRAGEN BCL Convert or DRAGEN Enrichment), the sequencing system might be able to store 4 flow cells worth of 25B data.

If the storage is not sufficient to start a run, an error message prompts you to clear space. For more information, refer to Clear Hard Drive Space.
