XLEAP-SBS Chemistry Quick Reference

This section provides an overview of the ways in which using the XLEAP-SBS chemistry impacts how users interact with the NextSeq 1000/2000 Sequencing System. To use XLEAP-SBS chemistry, there are compatibility requirements and procedural changes that must be followed. Those compatibility requirements and procedural changes are outlined here. The following software updates are customer-installable. For installation instructions for both online and offline installations, refer to the NextSeq 1000/2000 Control Software v1.7 Customer Release Notes on the Illumina Support Site.

Compatibility Requirements

Software

NextSeq 1000/2000 Control Software v1.7 or higher. Software installer and installation instructions are on the Software Downloads page of the Illumina Support Site.
DRAGEN v4.2 or higher. Software installer and installation instructions are on the Software Downloads page of the Illumina Support Site.

NextSeq 1000/2000 Control Software v1.7 is only compatible with DRAGEN v4.2. Install both software updates at the same time. After they are installed, these software updates cannot be reverted.

Reference Genome

DRAGEN v4.2 v9 Reference Genome. Download from the Product Files page on the Illumina Support Site.

Procedural Changes

Loading concentrations and cycle counts have changed. Refer to XLEAP-SBS Sequencing Considerations.
The required thaw times of XLEAP-SBS consumables are different. Refer to Thaw XLEAP-SBS Consumables.
Onboard denature and dilute procedures have changed. Refer to Denature and Dilute Protocol Generator.
Cartridge preparation now includes a step to reduce air bubbles and stabilize cartridge components, and inversion is done slowly. Refer to Prepare the Cartridge.
Q-table binning is updated to reflect the improved sequencing quality and accuracy of XLEAP-SBS chemistry. Refer to Real-Time Analysis Workflow.
